High-throughput Screening (HTS)

The Genomics Shared Resource is equipped with instrumentation, robotic automation, and software for running high-throughput screens using a variety of assay technologies. The facility is highly flexible, making it possible to run assays of any size using 96- and 384-well plates.

Automated Liquid Handling: A variety of liquid handling and robotic platforms make it possible to perform assays of any scale accurately and efficiently. We perform extensive quality assessment before every assay to ensure that all liquid handling is carefully calibrated and functioning correctly. We also perform ongoing quality control throughout the assay to ensure that conditions remain consistent across multiple plates, maintaining a highly controlled experimental environment.

Microplate Reading Technologies: The facility is equipped with two primary plate reading systems. These can be integrated into automated screening procedures or can be used as standalone plate reading units. Members of the Columbia University research community can schedule time to use our plate readers even if you are not conducting a high-throughput screening assay with us. Please contact us in advance to schedule a time.

High-throughput High-content Microscopy (HCM): High-quality images and high-content image analysis software are combined with high-throughput technologies for analyzing the results of high-throughput screening assays. The system available in the facility allows automated imaging and analysis of samples in a 96- or 384-well format.

Screening Libraries: We utilize an extensive catalog of small molecules for pursuing molecular screening including siRNA, shRNA, cDNA, and chemical probes.

Informatics: The Genomics Shared Resource uses high-quality informatics tools for accessing data processing protocols. Through a software platform called Pipeline Pilot (Accelrys), our clients can view and investigate the results of their screening assays through an intuitive user interface that can be accessed from any desktop computer. The software includes data visualization tools and other plug-in modules that can help you to explore, report, and share results. It also includes a specialized relational database for storing chemical and biological materials. Members of our staff can provide you with training and technical support to help you to utilize the tools that Pipeline Pilot offers.
